Description
Endoproteinase Lys-C is a serine protease specific for cleavage of amide and peptide ester bonds at the carboxylic side of lysine and S-aminoethylcysteine residues.
The recombinant Endoproteinase Lys-C is genetically engineered from the Achromobacter lyticus lysyl endoproteinase. It is expressed in E.coli and purified by high pressure liquid chromatography.
Recommended usage
The recommended ratio to the sample protein is 1:1000 to 1:20000 (w/w), the optimum pH is pH 7 - 9.5. The enzymatic reaction should be carried out at 30 - 37°C for 2 - 24 hours.
The use of enzyme inhibitors (such as aprotinin, DFP, leupeptin, and TLCK) or protein denaturation with guanidine chloride (GuCl) is not recommended.
Recombinant Lys-C is compatible with the following agents: 0.2% SDS, 1.0% Triton, 6 M Urea, 0.5 M NaCl, and 20% Acetonitrile.
Storage and stability
Store recombinant carboxypeptidase B lyophilized powder in a sealed container at 2-8°C. It is stable for 12 months.
The lyophilized powder can be reconstituted with ddH2O to the working concentration; and the solution is stable at 2-8°C for 7 days.
It is also recommended to dissolve the lyophilized powder in 25 mM Tris-HCl (pH 8.5) and make a stock concentrate solution. Aliquots of the stock solution may be stored at -20°C for up to 6 months. Avoid freezing-and-thawing cycles.
